Howard Grand Canyon University: LDR 800 February 22, 2012 INTRODUCTION This paper compares three empirical articles about ethical leadership practices that relate to the use of ethical power, authority, persuasion, and/or motivation. Article one was entitled â€Å"The relationship between ethical leadership and core job characteristics† and written by Ronald F. Piccolo, Rebecca Greenbaum, Deanne N. Den Hartog, and Robert Folger (2010). The second article was entitled â€Å"Impact of Ethical Leadership and Leader-Member Exchange on Whistle Blowing:†¦show more content†¦Article one participants were 174 junior and senior level undergraduate students and 107 masters of business education students from a large southeastern university in the United States. Article two consisted on 81 students from different postgraduate programs of a top engineering institute in India. Article three consisted of 191 working adults with a large university. Results of study The results of the articles were different. Article one resu lt showed that ethical leadership had a positive effect on follower effort and was mediated by task significance. The results also showed the relationship between subordinates’ job performance and ethical leadership was mediated by effort and task significance. The results of article two showed that whistle blowing is impacted strongly by ethical leadership behavior. Results showed that moral intensity of issues was moderated by the relationship between leader-member exchange (LMX) and ethical leadership as related to whistle blowing. Article three results found that deviance was negatively related to ethical leadership while follower organizational behaviors (OCB) were positively related to ethical leadership and were moderated by follower self-esteem. The results also found that ethical leadership should be used as a method to enhance citizenship and promote follower citizenship behaviors. First our spider revisits each site at least once every quarter to verify the information. Unlike other services software that just check if a link is â€Å"live,† we can tell if the site is under contraction, or the domain name is for sale, or other signs that the business is unavailable although the link remains active. More importantly, we e-mail the business at least once a year for changes, and reward companies that respond with a higher ranking. This gives us a good repsonse rate and ensures a proactive interest from each company to keep their data up-to-date. The primary targets are those established portal and navigation sites online that have a need to deliver this content. These include: Web-based directories and search engines, such as yahoo, excite, lycos Business directories and smaller search portals Industry-related sites portals (apparel.net, lawyers.com, etc.) Media sites (newspapers, television stations, radio) Direct mail services mailing list providers Content providers other infomediaries Together I have databased 500,000 web sites so far, and many of them are businesses in these categories. Each listing includes a link, business name, address, phone numbers, and email. For our meeting next week, I will provide an Access database of all the companies I have categorized in these fields. Would they be willing to pay to use our current and complete database at their site? Who else would be willing to use this data? What other types of businesses have I†ve missed? For an example to goal #3 above, it takes a lot of time energy for a local portal to gather the links of businesses in their area. They only catch the ones that are submitted to them, or the ones they find using their human editors. Likewise, a site focusing on the apparel industry can use a lot of energy effort to record and display links to clothing fashion sites in its industry. On the maintenance side, it can be time consuming to categorize their listings, check them to make sure they are current, and modify/delete old dead links. We want to partner with each site and provide the infomediary to exchange data: since they both share data with Speedy Pete†s database, they have access to not only our spider†s adds, but the other site†s listings, as well. The local portal may have listings related to the clothing industry that the apparel portal does not have and vice versa. Eutahnasia Essay Euthanasia is an issue that has been debated on many occasions. Both ethical and political questions have been raised. It is the one issue that can touch anyone, even politicians. Euthanasia is an issue that is controversial from all sides and is such a deeply felt issue across society. Parliament has a duty to address euthanasia. Terminally ill patients should have the opportunity of having doctor-assisted suicide or euthanasia as a legal option. Euthanasia should be legal so patients can die with dignity. It’s their right to choose when they want to die, and caring for such a terminally ill family member becomes a burden to the rest of the family. Many terminally ill sufferers want to have the choice to die with dignity and pride. By doing so they feel a sense of happiness that their suffering is over. Death with dignity basically means dying while still having some self-respect. Euthanasia is better then seeing patients not eating or drinking, consuming large amounts of pai nkillers and essentially being allowed to slip away. Patients also feel a sense of worth and importance in the society. Euthanasia has the potential to effect each and everyone of use; therefore, we have the right to choose how and when we want to die. One has to ask themselves if they were in the condition where they could not bathe, they could not go to the washroom, where they knew death was around the corner and the only thing keeping them alive was drugs, what would they do? If one cannot give consent to their death then whose body is it? Who owns their life? The decision is left up the Parliament who cannot feel what the patient is going through, how they are suffering, and their agony.Victims of a deadly virus have the necessary means to leave when they are ready and should not be faced with any higher authority telling them that it is against the law. Patients feel lose of control over their lives. Having a family member on their deathbed at times can become a burden on them . They could transfer them to an institution, however, that can prompt an ill person to take their life alone. When terminally ill patients can not take matters into their own hands, it places an agonising responsibility on the shoulders of the family. Family members should not be forced to choose between respecting the wishes of a loved one and obeying the law. Family members are motivated solely by their love and compassion for whom ever is ill and the need, at least in their mind, is that they should not suffer any more pain. Euthanasia is like a compassionate homicide. When one takes their own life, they are hurting no one but yet they are taking away all the pain they have suffered. Terminally ill patients fear the fact of being alive but not really living, mentally. Providing euthanasia as a legal option allows patients to die with dignity, to have the right to die, and to lessen the burden on family members. Sartre`s Existentialism Jean-Paul Sartre . . . the name is one of the most popular in modern philosophy. But who was he? What did he write and what were his works about? What was his role with regard to Existentialism? What is Existentialism, really? What life influences affected the person as whom he became famous? How would Sartre assess various social topics that we face today? What are the problems with Sartre's view of Existentialism and existence in general? These are the questions addressed in the following pages of this brief dissertation. His life Upon reviewing several sources, it is apparent that Sartre was a very disorganized and inconsistent individual. Sartre was obsessed with his intellect to the point of abandon of all else in his life - personal hygiene, honesty, organization, thoroughness, and more. It seems that he felt he was of superior intelligence in comparison to all others who surrounded him. He was not necessarily a great and original thinker, but rather a superb media sensation of sorts. Rather than developing Existentialist thought, he merely promoted it to amazing popularity through his eccentric lifestyle. Although he is best known for his association with Existentialism, it is interesting to note that he denounced its principles later in life and adopted Marxism, which he also later denounced. Jean-Paul-Charles-Aymard Sartre was born in Paris on June 21, 1905, the only child of Anne-Marie Schweitzer Sartre and Jean-Baptiste Sartre. Anne-Marie was the first cousin of Albert Schweitzer, the famed Nobel Peace Prize recipient, and the daughter of Karl Schweitzer, who had published texts on religion, philosophy, and languages. Jean-Baptiste was the son of Eymard Sartre, a doctor who had written several medical texts. Although his philosophy would deny it, it may have been fated that Jean-Paul would himself become a famous writer; it was in his genes. Jean-Baptiste and Anne-Marie were deeply in love. They married on May 5, 1904. Jean-Baptiste was enlisted in the French Navy and was away on an assignment when Jean-Paul was born 13 months later. Sadly, hardly more than a year after the birth of their son, Jean-Baptiste had returned home from China in November only to fall ill in March and die on September 17, 1906. After the death of Jean-Baptiste, Anne-Marie moved herself and her young son into her father's house, the Schweitzer home. Karl Schweitzer was a strict and domineering man and the year the two spent living there affected Sartre's life forever. His mother kept his hair long and dressed him in effeminate clothing, probably as a means of escaping the oppressive nature of her father. Schweitzer, however, disgusted by the child's appearance, took him to the barber one day and had his hair cut. Jean-Paul's ugliness then became apparent. Without the cloak of long hair and frilly clothes, his short stature, one eye that looked askance (from a juvenile illness), and awkward appearance were undeniable, even to his mother. He was ostracized by other children for his appearance. He was an outcast. At the age of eight he began to write scripts when he received puppets from his mom. Children tolerated him in order to be entertained by his shows. He basked in the attention. He began a pattern of outrageous behavior that it seems he believed would earn him popularity. Apparently it worked. In October of 1913, Eymard Sartre died and Jean-Paul fell under near complete control of the Schweitzers. When war broke out in 1914, it fascinated Sartre, and he wrote some short stories about it. In 1915 Jean-Paul was enrolled at Lycee Henri IV, a highly regarded school. Here he found children he could relate to: intellectually stimulating and of his class level, children who could respect him for himself. Yet, even at this early age, it was apparent to his teachers that Jean-Paul did not hone any of his thoughts; his intelligence was apparent, but he merely skimmed over many subjects without delving into any in depth. His mother remarried when he was twelve, to the apparent disapproval of Jean-Paul. The new family moved to LaRochelle in 1917, but after Sartre got into trouble on several occasions, he was returned to Lycee Henri IV where he was a boarding student. At this time he became close with Paul-Yves Nizan, a quiet and shy boy of considerable intellect. Where Sartre was disorganized, slovenly, and incomplete, Nizan was orderly, stylish, and thorough. Nizan was prone to fits of depression and drinking, to the fascination of Sartre. The two were nearly inseparable throughout college and beyond. In 1922 the two enrolled

Differentiating Fire Ants from Other Ant Species Look for the following traits to identify red imported fire ants: Nodes: Fire ants, whether native or imported, have two nodes at the constricted waist between the thorax and abdomen.Antennal clubs: The antennae of fire ants (genus Solenopsis) consist of 10 segments, with a two-segment club.Small size: Red imported fire ant workers measure just 1.5 mm to 4 mm.Size variation: Red imported fire ant workers vary in size according to caste.Color: Red imported fire ants are reddish brown, and the abdomen is darker than the rest of the body.Standard proportion: The heads of red imported fire ants will never be wider than their abdomens in any worker caste. They resemble the piles left behind by digging gophers.Mounds usually appear in spring or fall, or after cool, wet weather when breeding conditions are best.Unlike those of native ants, red imported fire ant mounds do not have an opening in the center. The ants enter the mound from tunnels below ground level.Red imported fire ant mounds usually measure up to 18 in diameter, but will often be considerably smaller.Fire ants build mounds in open, sunny locations.When the mound is disturbed, the white brood will be visible. The larvae and pupae may look like grains of white rice in the soil. Fire Ant Behavior Fire ants are the hotheads of the ant world. You may be able to identify fire ants by observing their behavior. Any disturbance of the nest will elicit a quick response, with dozens of fire ant workers storming from the nest to do battle.Fire ants will typically climb vertical surfaces when disturbed. The author defines poverty as the dearth of the necessary resources that enable the people to meet their basic needs and extends the commensurate statistics regarding the urban, suburban and rural incidence of poverty. The author does support the extension of government aid to the deserving, while taciturnly mentioning that many people suffer from poverty because they are lazy. The writer also blames the indulgence in substance abuse like tobacco and alcohol as the reason as to why the poor remain poor. Yet, the writer again contradicts one’s assertion by saying that a great number of the poor receiving the government aid do tend to be employed. In the end, the writer conclusively asserts that the government should spend money on imparting skills and education to the poor, rather than wasting the taxpayers’ money on extending nutrition, health and housing. Overall the approach of the writer seems to be discernible against the government funding of the poverty alleviation programs. The poor tend to remain poor because they simply do not have a place in the national financial chain and many times, as said by the author, they lack the skills, education and connections to be able to fit into the national financial dynamics. Hence the government spending on the poverty alleviation programs is the most efficient way to make way for the allocation of the national wealth in favor of the impoverished segments of the society. The government has a self-interest in doing so in the sense that allowing the poverty to rise beyond a certain limit could lead to an unstable society marred by political unrest and crime (Lal & Myint 8). It is rightly said that the hungry minds are angry minds. Hence, state expenditure in poverty alleviation programs allows the government to channelize the skills, abilities and energy of poor sections of the society towards self-development and nation-building. State sponsoring of the poverty alleviation programs is not a choice but an important n ecessity and requisite. Hence, there is no scope or possibility for casting doubts on the relevance or necessity of the government spending on the poor or poverty alleviation programs. The writer himself conveyed that a majority of the poor people receiving government help does have jobs and they tend to work. Hence, the tongue in cheek assertion that the poor people are poor because they happen to be lazy is utterly nonsensical and biased. Poor people, in fact, happen to be poor because they do not have a say in the national and international policy-making procedures and frameworks, where they could chalk out the apt policies that would help them have a share in the process of wealth generation and allocation (Global Issues 1).

The capital of one company could be handled well if they have the right organizational strategies. In electronic business the common problems that may arise will be rejection of service attacks, viruses, obliteration, and theft of their intellectual property. These problems could be prevented if the organization has good managerial considerations and technical options. There should be a balance between internal and external security management (Greiger and Wegman., 2002). Each component contributes to the behavior of the system. No component has an independent effect on the system. The behavior of the system is changed if any component is removed or changed. The system has an outside an environment which provides inputs into the system and receives outputs from the system. All customers want a company that would work to their advantage without any surprises regarding deals made between him and the financial company. They would love someone who could provide them and first-rate service and a bank that has security that they could provide service for a long time. Money matter is one issue among the working citizens that requires them to think a hundred times before getting the service of any financial company. There are a lot of companies that tries to persuade them to invest in their company and that they will be getting their money’s value. Assessing one program’s effectiveness is crucial to its success. Evaluations of the advertisements and marketing strategy could guide the company if there adjustments are needed. Evaluation of the trend of existing customers would be one way of checking if objectives are being met. Bill Gates :: essays research papers

In his high school years, Bill Gates proclaimed that one day he would be a millionaire. He greatly underestimated himself. He became interested in computers while in the 8th grade. His school received a hookup to a high powered computer. Bill and his good friend Paul Allen used to skip gym class to use the computer. Sometimes they would even break into the school at night to spend time on the computer. Eventually Bill made his first program. A virtual tic-tac-toe game. The Computer Center Corporation (CCC) wanted to rent a computer from the Digital Co., but they couldn't pay for it. Digital said they would provide it at no cost if they could find any bugs in their computer. CCC put an add out in the local paper, and Bill and Paul applied immediately. Although skeptical about hiring two teenagers to do the job, their doubts were quickly dissolved. When they were done, Bill and Paul had compiled a list of bugs that was over 300 pages long! Bill was next hired by his school. They wanted him to write a program for use in class scheduling. Bill was happy to do it. He used a programming language called FORTRAN for the first time in this program. FORTRAN was one of the major languages later used at Microsoft. With this program, Bill first took advantage of his skills. He designed the program to put him in classes with no other boys and all the girls he thought were cute, what a guy! At this time Paul and Bill also started their first company called Traf-O-Data. Keep in mind that they are still in high school! They designed a program that could analyze traffic counts by machine so counties could get government funding for road projects. They had clients in British Colombia and Delaware but went out of business in 1973 when the government offered the same services for no charge. Bill Gates scored a perfect 800 on the math portion of the SAT's. However, instead of going into computers, which would seem logical, he went to Harvard to become a lawyer. In 1974, a company in Albuquerque, NM, known as MITS made the first home computer. The computer was useless because it had no monitor or keyboard. In place, it had switches to input data and a device similar to a printer to display data. This computer was called Altair and when Bill and Paul heard about it, they were a-twitter.

Personal Finance Essay

Without financial knowledge, a budget and/or a financial plan in place you can have an income of 100,000 thousand dollars a year and still be living paycheck to paycheck and eventually end up in financial ruin. If a budget is not in place it is very likely to spend more money than you are making. By accounting for all income you receive and spend you are able see you’re spending habits. To assess whether you’re spending habits are good or bad, need to be improved or adjusted. For instance my family loves to eat out. If I noticed that my family spends $400 dollars a month on fast food/eating out then I need to decide what’s essential to my household and what’s not; if spending that amount on fast food is not essential then I need to make a conscience effort to eat at home more and decide on an acceptable amount and give my family a fast food budget, a certain amount for fast food monthly. I can also add that additional money not spent on eating out to our savings account or add it to a 401k, IRA, or even pay extra on our credit cards. By making good personal finance choices money can be saved monthly to add to retirement income. Personal finance choices affect your life now but most importantly they will affect how comfortable you are able to retire. To design a budget for earnings, spending, saving and investing a financial plan is a necessity. To create a financial plan for earnings, spending, saving and investing you must first understand these things separately, how they all work together and affect each other and that they are a necessity to laying a solid foundation for your family’s future. First, a detailed budget is needed for a clear picture to see how much money you have, what you spend your money on, how much you’re spending and the amount left over. A budget allows you to make sure your money is going where it should. After a budget is created and you’ve cut back on the little things that aren’t essential and you’ve focused on getting out of debt by not borrowing more than you can afford to pay back and paying more than just the minimum on credit payments then it’s time to focus on a plan for saving and investing. It is important to take advantage of tax – advantaged accounts like employer 401k plans, individual retirement accounts and special retirement accounts for the self employed. These accounts give way for credits, tax deductions and tax free earning on retirement savings. Insurance is also an important part of this process to protect your life, your capability to earn income and the roof over your head.

Definition of od interventions: An OD intervention can be defined as â€Å"the set of structured activities in which selected organization units engage with a task or a sequence of tasks where the task goals are related directly or indirectly to organizational improvement † â€Å"A set of sequenced and planned actions or events intended to help the organisation increase its effectiveness†. In an od intervention the entire process of diagnosis, alternative generation and making action choices are jointly conducted and od will also examine the process of such diagnosis, action planning and implementation.Criteria for Effective Interventions: In OD three major criteria define the effectiveness of an intervention: 1. The Extent to Which it (the Intervention) fits the needs of the organization. 2. The degree to which it is based on causal knowledge of intended outcomes 3. The extent to which the OD intervention transfers change-management competence to organization members. Characteristics of od intervention: First An od intervention will focus on the organizational processes apart from the substantive content of an activity.Secondly an od intervention would generally focus on a work team as the unit of analysis and change towards effective behaviour Thirdly od would normally view change as an on-going process and would rely on a collaborative management of work culture Types of od interventions : These interventions can be clubbed into twelve groups and they are diagnostic, team-building, inter-group activities, survey feedback methods, education and training programmes, techno-structural activities, process consultation, the management grid, meditation and negotiation activities, coaching and counseling and planning and goal setting activities. ) Survey Feedback: It recounts to an approach survey through well planned assessment or Interviews. It covers the feedback to the customer organisation. It has a demand and significance in a genuine situation. The attitude survey can have a purpose to measure the process; for instance, communication, decision making and leadership at different levels. The data produced under this system is perceptual and attitudinal.. A review of the product is prepared in a group discussion.Generally feedback of results is specific to the group which prepares the data. As a result strategies are planned to resolve the organisational problems. Consequently, the process of survey feed back includes: collection of data, feedback, development of action plan and follow-up. D and involves the process of educating the concerned persons about M. B. O, agreement upon clear cut quantifiable objectives, evaluation of objectives and feedback for deviation and coercive action. . M. B. O is wide-ranging overall managerial philosophies that focuses upon joint goal setting and synthesize the individual’s goal to organisational goals.The entire organization will have the feeling of unity as all levels of management of an organization are involved in goal setting. The programmes relating to M. B. O can be effective if they are properly implemented and needs the support of the top management. 7) Process Consultation: It can be described as the set of activities on the part of the consultant that helps the client to perceive, understand and act upon the process events that take place in the client’s environment. It is not harsh on our environment making it a safe and wise choice. There is no pollution, noise, or hazardous waste as a result of this conversion of the sun’s energy. The electricity created is clean, reliable, and renewable. The use of Solar Energy also decreases our dependence on fossil fuel and other energy sources much less friendly on our environment by reducing greenhouse gases. Solar power systems are cost efficient and provide it’s users with fixed energy costs. As the use for this â€Å"clean energy† increases so does the development opportunities and local jobs. The use of heat and light simultaneously obtained from the sun is a brilliant and beneficial asset to us on many different levels. How we use Solar Energy In order to utilize the sun’s energy, there must be some type of conversion for channeling. This device is called Photovoltaic (PV devices). It can also be referred to as â€Å"solar cells† which transform sunlight into direct electricity. These PV cells are individually grouped into sections which are used in a vast array of applications sometimes ranging from small single cells that are capable of charging small watches, to larger, more complex systems that are capable of powering homes and even expanding to power plants that may cover many acres of land. Photovoltaic is a method used to generate electricity through solar radiation while transforming direct electricity by using semiconductors which display photovoltaic effect. This power generation uses solar powered panels consisting of numerous solar cells of photovoltaic material. There are several materials used to make photovoltaics including copper indium gallium, polycrystalline silicon, cadmium telluride, m onocrystalline silicon, and selenide/sulfide. As the demand for renewable energy increases, the development of photovoltaic systems and solar cells made astounding advancement over the past few years. Photovoltaic systems or PV’s rely on solar panels in order to change light from the sun into usable electricity. â€Å"This system is made up of one or more photovoltaic (PV) panels, a DC/AC power converter (also known as an inverter), a racking system that holds the solar panels, electrical interconnections, and mounting for other components. Ying-ying asked what a â€Å"secret wish† was, and the answer was that is was on that you cannot ask. Ying-ying was young as this point, so she asked why. Amah’s response to Ying-ying was, â€Å"This is because?because if you ask it?it is no longer a wish but a selfish desire?Haven’t I taught you-that it is wrong to think of our own needs? A girl can never ask, only listen.† Here she is telling that by making a wish for herself she is being selfish and forgetting that she is not the important one, but the good of the whole is the important one. Amah is als o hinting at obedience when she tells her daughter that she is not to ask questions, but just listen to the men and go along with it. Arranged marriages are a very tough thing to go through. Marrying someone because one has to has to be hard because one is not marrying out of love. This is exactly the case in â€Å"Waiting for Mr. Kim.† Gracie two twin sisters have already run away from to and eloped with two men. Their marriages were not arranged because they left before their father could arrange them. Now, with her sisters gone, Gracie is the only remaining daughter, and she is somewhat frowned upon because she was the â€Å"third daughter.† She did not really like the two men that her father was thinking of arranging the marriage with, but it did not matter. It did not matter because as her mother told her, â€Å"Girls don’t choose.† The girls just sit there and look pretty while their fathers choose for them, they have to comply with their fatherâ₠¬â„¢s decision. They are obedient, and at the same time respectful of the decision that is made. Another characteristic that appears in this story is that women made to think that they are second class compared to men. When her Mr. Kang, Gracie’s father, would walk, he would sometimes reach back to hold Mrs. Kang’s hand, but she would pull away. Not only would she pull away, but she would â€Å"stay behind as she cleaned her purse or took forever with her coat, just to have it the way she had learned it, her husband a few places ahead, women behind.† Her mother not only believes that this is how women should act and it is a part of their femininity, but she is passing this way of thinking onto her daughter. Her mother is relaying the message to her daughter that no matter what you need to show your husband respect and let him know that he is in charge. Both of these stories have displayed that there are certain characteristics of females and they are inner stren gth, the good of the whole is better than the good of the individual, find things out for oneself, honor and respect, and obedience. All of these characteristics have displayed not only what it means to be feminine, but also what it means to be female. It is not so much an Asian female, but it is because that is how their mothers were raised and that is how their mothers will raise them. There are really no American females for the daughters, or mothers, to see and mimic. The only way of life that the mothers know is the Asian way of life, therefore that is the only way that can be passed down onto their daughters. Sometimes that is hard for the daughters to understand because the daughters have grown up in American culture, and they know what it is like to be an American woman. No matter how one views this situation, these stories focus on what it means to an Asian female, but more generally what the mothers know best on how to teach their daughters on how to be female.

Proto-Oncogene And Tumour Suppressor Gene Protooncogene is found to be normally expressed in all vertebrates including humans. It is also found to be expressed in some insects and yeasts. Protooncogenes play an important role regulating normal cell growth and differentiation. They also perform functions such as signal transduction and mitogenic signal execution RAS, MYC, TRK, ERK are some examples of protooncogenes (Novakofski 1991). Oncogenes are derived from protooncogenes during the process of carcinogenesis. The formation of active oncogene leads to abnormal cell proliferation and thus contributes to the formation of tumour (Alitalo and Schwab 1986). A protooncogene can be converted into an oncogene in a variety of ways such as (i)Transduction induced by retroviruses which cause the integration viral DNA with the host. This DNA on translation give rise to viral proteins which cause the activation of protooncogene, thereby leading to cancer. (ii) Mutation within the protooncogene leading to an increased activity of the pr otein. (iii) chromosomal translocation where the gene for proto oncogene is translocated to other loci, which cause its abnormal expression. For eg in the translocation of abl in chromosome 9 to bcr region chromosome 22 as found in Philadelphia chromosome (Rowley 1973) (iv) Amplification induced by gene duplication, mis-regulation of gene which leads to overexpression of protooncogenes. (Todd and Munger 1999) Unlike oncogenes, the tumour suppressor genes normally inhibit cell growth in variety of ways such as (i) repressing the genes important for cell cycle progression. For eg., pRb (retinoblastoma ) controls the G1 stage by being hypophosphoryated and not allowing the release of transcription factor E2F which promotes G1-S transition. (ii) Stopping cell cycle on detecting DNA damage. DNA damage induces p53 which now leaves from its interacting partner Mdm2 and increases the activity of p21. This p21 protein then inactivates cyclin dependent kinases which are essential for cell cycle progression, (iii) Apoptosis may also be promoted by p53 when damage is irreversible. (iv) inducing DNA repair proteins to repair DNA damage and prevent abnormal cell proliferation. (Yoshida, et al. 2000) There are a number of mechanisms by which a cancer cell survives in a body. This is illustrated in the diagram below.(Weinberg and Hanahan, 2000) Treatment The choice of therapy depends upon the location and stage / grade of the tumour, as well as the health of the patient. There are a number of therapies for treating cancer such as surgery, chemotherapy, radiaotherapy, hormonal therapy etc., The deregulation of many genes in cancer cells leads to the over-expression of altered proteins which can be used as biomarker which can provide help for the diagnosis and / prognosis of the treatment but can also be used as potential target for immunotherapy. Cancer antigens and their classification Tumour antigen is an antigenic substance produced in the tumour cells and triggers an immune reaction in the host. They are useful in identifying tumour cells and are used in cancer therapy. Tumour antigens are classified as Cancer testis antigen, Differentiation antigens, Tumour specific unique antigens, Overexpressed Self Antigens, Viral antigens, Post-translationally, Oncofoetal antigens, Idiotypic Antigens as shown in the table below Copied from (Li, et al. 2005) Amongst all these categories, cancer testis antigen represent the most promising group of antigen to be used in future immunotherapeutic interventions due to their restricted expression to mainly to tumour cells with the exception of placenta and testis which are immune-privileged sites and therefore would not pose any real issue for the risk of autoimmunity.(Simpson, et al. 2005) HAGE HAGE also known as DDX43 and CT13 antigen belongs to this category of antigens and was first identified by Martelange et al using cDNA subtraction approach of a human sarcoma cell line.(Martelange, et al. 2000)Using rational hybrid analysis, it was found that HAGE gene is located on chromosome 6. HAGE was found to be overexpressed in several tumours. There is a low expression of HAGE in normal tissues since the expression is regulated by hypermethylation HAGE is also over expressed in tissues such as testis, placenta, and ovaries. HAGE is usually expressed at the m-RNA level though recent studies in melanoma cells suggest that HAGE could also be expressed at the protein level.(Mathieu, et al. 2010) The name DDX43 comes from the fact that HAGE also belongs to another group of protein known as Dead box proteins and referred to as DDX43. (Abdelhaleem 2004) Dead box proteins These proteins were discovered in the late 1980s. (Gorbalenya, et al. 1989)Dead box proteins belong to the family of RNA helicases and play an important role in the transcription, post transcriptional modifications such as splicing, transport, translation, decay and biogenesis of ribosomes. NTP hydrolysis provides energy to these enzymes and they unwind dsRNA or disrupt RNA-Protein interaction. DDX and DHX are widely studied Dead Box Proteins. There are many classes of DDX and HAGE belongs to DDX43. They are so named because they contain the amino acids D-E-A-D (Asp-Glu-Ala-Asp) in one of their motifs namely Motif II. Besides they also contain motifs such as motif I, Q-motif, motif VI, motif Ia, Ib, III, IV and V as shown in the diagram below. These motifs bestow properties such as ATP binding and hydrolysis, RNA interaction, remodelling activity etc to these proteins. The consensus sequence of DEAD box family is shown below Copied from (Linder 2006) Regulation and Function of HAGE Both function and regulation of HAGE have not been studied in great detail (Scanlan, Simpson and Old 2004). In normal tissues HAGE expression is regulated by DNA hypermethylation, Histone modifications such as histone aceylation, histone deacylation and histone methylation. However in cancer, there could be many mechanisms of upregulation of HAGE. The most commonly thought mechanism is the demethylation of DNA which leads to HAGE over-expression. (Roman-Gomez, et al. 2007) HAGE could also be induced due to transformation of normal cells during cancer or due to the action of oncogenes. HAGE might also be induced randomly. HAGE may play a role in pre-mRNA splicing, ribosome biogenesis, transcription and initiation of translation. (Rocak and Linder 2004) This project will focus on Head and Neck Cancer. Head and Neck Cancer Head and Neck cancer are the cancers that arise from the upper aerodigestive tract such as nasal cavity, oral cavity, pharynx, larynx and paranasal sinuses. Most of them are of squamous cell carcinoma type of the head and neck and have been given the acronym -SCCHN or HNSCC. (Argiris, et al. 2008). The diagram of the anatomy of the head and neck is illustrated below Copied from (Wu, et al. 2009) More than half a million patients are diagnosed with head and neck cancer every year. Head and Neck cancer in fact accounts for more than 3-10% of the cancers(Gourin and McMains 2005). Smoking, alcohol, chewing of betel leaf with tobacco and areca are the major risk factors for this cancer. Recently, HPV type 16 has been identified as one of the causes for head and neck cancer and account for 40% (Goodger and McGurk 2000) The progression of this tumour is complicated. It progresses from the normal features to hyperplasia, mild dysplasia, moderate dysplasia, severe dysplasia, carcinoma, invasive carcinoma and metastasis. The progression may be caused by due to genetic instabilities such as loss of heterozygosity of chromosome 9p21, inactivation of of p16 and loss of 3p, loss of 18q, inactivation of PTEN, some translocations, 17Beta heterozygosity or TP53 mutation. (Argiris, et al. 2008) Copied from (Argiris, et al. 2008) Interestingly HAGE was found the John van Geest group to be overexpressed in 40% of HNSCC cancers (unpublished data) and although much of its function is now emerging no information exist at the moment regarding the regulation of its expression. However it has been observed that cells left for longer period of time in the incubator had a significantly higher expression of HAGE at the mRNA and protein level. Aim of this project The observation that HAGE was over-expressed in 40% of HNSCC led to hypothesis that cells under stress i.e lack of nutrients, decrease oxygen level, as well as other factors influence HAGE expression. Moreover it has been found that HAGE is also expressed in 20% of Acute Myeloid Leukemia and 50% Chronic Myeloid Leukemia as well as many solid tumours such as HNSCC (Adams, et al. 2002). This suggests that there must be something in common between these 2 very different forms of cancer. CML and solid tumours are known to have a high expression of reactive oxygen species (ROS), therefore it might be possible that ROS induces HAGE expression. Thus this project will focus on the effect of oxidative stress has on HAGE expression. The effect of other stress such as temperature may also be investigated. METHODS Cell Culture The PCI 13 and PCI 30 cell lines would be supplied by Prof E Tatour. These cell lines are specific for Head and Neck cancer. The cells would be grown in RPMI 1640 +10%(v/v) FCS+2mM L-glutamine and incubated at 37oC and 5%C02 atmosphere. Extraction of m-RNA The culture media would be removed and washed with DPBS(Dulbeccos phosphate buffer saline). Then DPBS will be removed followed by the addition of RNA-STAT 60. RNA pellets would be then retrieved and re-suspended in ddH2O. RNA extracted is then quantified using NANODROP 8000 UV spectrophotometer. RNA would be then added in measurement wells of the spectrophotometer. The spectrophotometer would be read at 260 and 280nm and the amount of RNA will be calculated. The RNA concentration is calculated in  µg/ µl before being adjusted to 1 µg/ µl by ddH2O. RNA samples would be then stored at -80oC and used for conducting RT-PCR and RTq-PCR in the future. RT-PCR (Reverse Transcriptase PCR) From the extracted RNA cDNA will be synthesized. 2  µg of RNA sample will be mixed with 1  µl of oligo-dT primers and diluted to 15  µl of ddH20 in an Eppendorf tube. All RNA samples would follow the same process. UNO Thermoblock will be used to heat the tubes to 70 °C for 5 min which allows the primer-RNA annealing. 10  µl of reverse transcriptase mix, which will be prepared by mixing Muloney Murine Leukemia Virus 5-X buffer, dNTPs, RNasin RNAse inhibitor, M-MLV-reverse transcriptase and ddH20. The tubes will be then pre-heated in a water bath at 39.2 °C for 80 min which allows cDNA synthesis. Tubes will be removed and kept again in the UNO-Thermoblock and heated to 95 °C for five minutes which stops the reaction. These tubes would be stored at -20 °C for future use. Real Time qPCR cDNA generated from RT PCR will be used as a template and the reagents used will be (i) 6.25 µl iQ Sybr Green, (ii) 0.5 µl gene specific sense primer (iii) 0.5 µl gene specific anti-sense primer, (iv) 4.75  µl ddH20 and 0.5 µl cDNA template. These reagent mixtures will be added to all tubes for a specific gene. Samples will be usually carried out in duplicate with a negative control that contains the reaction mixture without cDNA. Rotogene 6000 real-time qPCR analyser will be used to carry out RT qPCR. Relative gene expression using 2ΔCT method will be calculated with the help of the expression of housekeeping genes HPRT-1, HSP-27 and HSP-90. The primers which will be used in real time qPCR experiment is given below. These primers are supplied by mwg-Eurofins. The primers have a stock concentration of 100pmol/ µl and will be diluted to a working concentration of 10pmol/ µl. From this working 0.5 µl (5pM) of primers will be used. Total Protein extraction Cells will be grown to 75% confluence in T75 flasks. Cells will be trypsinised, washed and re-suspended in DPBS and these cells will be then counted using trypan blue. Cells will be diluted to 1X106/ml using DPBS and 5 X106 cells would be taken in a 1.5ml eppendorf. Centrifugation will be done to pellet the cells. The supernatant will be discarded and lysis buffer containing a cocktail of RIPA buffer and 10% protease inhibitor will be added to the pellet. The eppendorf tube will be then placed on a tube rotator at 4 °C for 30 min. It will be then kept on ice for 30 min. Cells will be again centrifuged at 14000 RPM for 30 min at 4 °C. Aliquot of the supernatant will be prepared to be stored at -20 °C for future use. Total Protein Assay Total protein assay will be carried out using BioRad Dc protein assay reagents. Series of dilutions (0.2,0.4,0.5,0.8,1.0,1.5,2.0mg/ml) will be created using stock BSA solution with the concentration of 10 mg/ml. Protein extracts and standards would be tested in triplicates and duplicates respectively. Assays will be performed in 96-well round bottom plates. 25 µl and 200  µl of reagent A and B will be added to all the samples and the samples will be incubated at room temperature for an hour. Proteins will be then read at 750nm and their concentrations would be calculated based on the standard values. Western Blotting 30  µl of sample will be loaded into the wells of SDS PAGE gels. 1X tris-glycine-SDS will be used as the running buffer. A known molecular weight ladder will also be run alongside the samples. Initially 70V current will be applied to aid the migration of proteins through the 5% stacking gel. Once the proteins reach the 10% separating gel, 90V current will be applied. After the proteins run through the gel, they will be transferred to the PVDF membrane with the help of liquid transfer. This membrane will be prepared by washing with 10% methanol for 5 seconds, ddH2O for 5 min and transfer buffer for 10 min. Liquid transfer will be carried out using the following steps. 1. Cold transfer buffer will be used to immerse a gel frame. To this gel frame a sponge pre soaked in transfer buffer will be placed Then a filter card, gel, PVDF membrane, another filter card and a second pre soaked buffer sponge will be placed in order starting from the first filter card. Proteins will be transferred with the help of electric current of 100V applied for one hour. After completion, membranes will be cut and treated with different antibodies.10% (w/v) Marvel milk solution will be used to wash the membrane for one hour at room temperature. This washing blocks the non-specific binding sites. The blocking solution will be then discarded and the antibody diluted to 10% (w/v) in Marvel milk solution will be added. This antibody coated membrane will be agitated overnight on a plate rocker at 4 °C. The next day there would be 3, 10 min washes with TSBT(Tris-Buffered Saline-Tween-20). During these washes the membrane vessel will be agitated in plate orbital shaker at room temperature. After completing the washing process, a secondary antibody specific to the primary antibody will be added. The steps for adding the secondary antibody are same as the primary antibody addition except for the fact that no overnight incubation is required and the secondary antibody will be incubated only for one hour while being agitated on the shaker at room temperature. The marker will also be stained using streptavidin-HRP secondary antibody. After performing the above step the membranes will be developed. Membrane development will be performed by placing the membrane in a tray and washing it with ECL reagent. The membrane would be then exposed for a certain period of time using CCD camera. Assay for ROS-DCFDA Stress Test Stock Solution Preparation The stock solution of Hanks Buffered Salt Solution (HBSS)will be prepared according to the manufacturers protocol. DCFDA(2,7-dchlorofluoroscein -diacetate) Assay To test ROS levels DCFDA test will be performed. Before conducting the DCFDA assay, the standardization of optimum concentrations of H2O2 and DCFDA for detecting cell stress should be performed. It is important to note that H2O2 should be added only sub-lethally and should not be added in proportion which may cause cell death. The PCI 13 and 30 cells will be plated out in two 24 well plates. 1ml of each type of cells plus RPM1640 10% FCS media will be added to 8 wells per plate and will be incubated overnight at 37 °C. The media will be removed the next day and increasing concentration of DCFDA will be added to both plates and the cells will be incubated with Tinfoil wrapping to prevent light exposure and incubated at 37oC for 30 min. The cells will be then washed and then cold HBSS will be added. After this step the cells will be stressed with increasing concentration of H2O2 (see diagram below). The cells will then be again wrapped in tin foil and placed on a rocking platform for 15 min. The same method will be followed for treating other wells with different concentrations of DCFDA. Experiments will be performed at least twice for each cell line. H2O2 will be removed after 15 min and cells will be trypsinised with 100microL trypsin and Versene. 800 microL of DPBS will be added to one of the 2 wells an d will be pipetted thoroughly so as to remove the cells from the well surface. This will be then transferred to the other well and then 1ml of solution would be transferred to Flow associated cell cytometry (FACS) tube for analysis. The same procedure will be carried for other sets of wells. DCFDA fluorescence would be measured using Gallios flow cytometer and results would be analysed in Kaluza program. Time Line For The Project During the month of May, all techniques such as Western Blotting, mRNA/protein extraction, Real Time PCR will be learnt. This time is indicated in red colour in the graph below. This will overlap with the time duration, from May to the end of June, during which hydrogen peroxide experiment will be performed as per written in the method section and the expression of HAGE will be monitored at both the mRNA and protein level. Also from the start of June to the end of July repeat experiments will be performed and if time permits, the effect of thermal stress on HAGE expression will also be investigated. This is indicated in the graph below. SUMMARY Cancer Testis Antigens (CTA) are antigens that are expressed in a variety of tumours. They are usually absent in normal tissues with the exception of testis and placenta where they are expressed as self tolerant antigens. Since these antigens have strong immunogenicity and their expression is mostly restricted to tumours, they are ideal targets for cancer immunotherapy. Therefore much research is ongoing for the identification of CTA. Recently helicase antigen HAGE was identified as a CTA and was found to be over-expressed in HSNSCC, AML and CML. Thus there might be a common link between these different types of cancers. Moreover ROS is known to be induced in all these tumours and it may be possible that ROS causes increased expression of HAGE. Thus the aim of this project would be to investigate the effect of oxidative stress on HAGE expression. HAGE expression would be analysed both at the m-RNA and protein level in two cell lines namely PCI 13 and PCI 30 using techniques such as m-RNA extraction, RT PCR, Western Blotting and Protein assay. The ROS level would be evaluated using the DCFDA assay. Since the mechanism of HAGE regulation is unknown, positive result in the project would help in elucidating a mechanism by which HAGE could be regulated.
